为了对黄鳍棘鲷增殖放流效果进行评估,本研究应用课题组构建的基因组开发了多态性高的微卫星分子标记,建立了黄鳍棘鲷的亲子鉴定技术,在此基础上,对2023-2024年厦门湾黄鳍棘鲷增殖放流效果进行了评估,具体研究结果如下:(1)多态性微卫星引物筛选。在黄鳍棘鲷基因组上随机挑选110个微卫星位点设计引物,以野生黄鳍棘鲷群体为材料初步筛选出9个多态性较高的位点。(2)亲子鉴定技术的建立。以系谱关系已知的黄鳍棘鲷家系及其亲本(亲鱼20尾,子代72尾)为材料,利用上述筛选到的9个微卫星位点进行亲子鉴定分析,结果显示:9个微卫星位点共检测到145个等位基因,平均等位基因数(Na)为16.11,平均观测度杂合度(Ho)为0.788,平均期望杂合度(He)为0.810,平均多态性信息含量(PIC)为0.785,具有丰富的多态性。运用Cervus 3.0.7软件对该群体进行分析,结果显示:在置信度为95%条件下,双亲性别已知模型的亲子鉴定成功率为98.61%,双亲性别未知模型的亲子鉴定成功率为97.22%。(3)增殖放流效果评估。对114尾放流亲本和264尾回捕个体利用上述建立的亲子鉴定技术进行分析,在回捕的264尾黄鳍棘鲷中,有8尾被确定为放流亲本的子代,即对野生黄鳍棘鲷群体的资源贡献率为3.03%。研究结果可为今后黄鳍棘鲷种质资源鉴定,增殖放流效果评估提供可靠的技术手段。

To evaluate the stock enhancement effectiveness of Acanthopagrus latus, this study employed highly polymorphic microsatellite markers developed by our research group using the species' genome and established a parentage assignment technology. Based on this, we assessed the stock enhancement effectiveness of A.latus in Xiamen Bay from 2023 to 2024. The specific findings are as follows: (1) Polymorphic Primer Screening: From 110 randomly selected microsatellite loci in the A.latus genome, we designed primers and preliminarily screened 9 loci with high polymorphism using a wild A.latus population. (2) Establishment of Parentage Assignment Technology: Using known pedigree families of A.latus, including 20 parental fish and 72 offspring, we performed parentage analysis with the 9 selected microsatellite loci. The results detected 145 alleles across the loci, with an average allele number (Na) of 16.11, an average observed heterozygosity (Ho) of 0.788, an average expected heterozygosity (He) of 0.810, and an average polymorphic information content (PIC) of 0.785, indicating high polymorphism. The analysis using Cervus3.0.7 software showed that under a 95% confidence level, the parentage assignment success rate was 98.61% when the sex of both parents was known, and 97.22% when the sex was unknown. (3) Stock Enhancement Evaluation: The parentage assignment technology was applied to 114 released parental fish and 264 recaptured individuals. Among the 264 recaptured A.latus, 8 were identified as offspring of the released parents, indicating a resource contribution rate of 3.03% to the wild A.latus population.The results provide reliable technical methods for future genetic resource identification and stock enhancement evaluation of A.latus.
