考氏鳍竺鲷(Pterapogon kauderni)早期色素细胞发育和体色变化

本研究以考氏鳍竺鲷(Pterapogon kauderni)为研究对象, 采用显微观察和拍照的方法记录了考氏鳍竺鲷早期发育过程中色素细胞的分布及形态变化以及体色变化过程。在水温(27±0.5)℃条件下, 考氏鳍竺鲷胚胎经15~22d孵化。受精后4d(4 days post fertilization, 4dpf)胚胎发育至器官形成期, 6dpf黑色素在眼球出现, 7dpf黄色素细胞在体节处出现, 8dpf虹彩颗粒反射小板在眼球出现, 19dpf白色素细胞在第二背鳍出现。考氏鳍竺鲷体表图案的形成自12dpf头部眼间隔黑色素细胞的出现开始, 21dpf吻部第一色带初显, 孵化后1d (1 day post hatching, 1dph)卵黄囊吸收完毕, 体侧四条色带与尾部Y形色带基本形成, 70dph身体中部第三和第四色带之间开始出现白色斑点, 220dph体色模式与2龄成鱼一致。研究结果可为探究考氏鳍竺鲷体色形成调控机制和观赏鱼进化学说的发展以及新品种的选育提供理论依据。

Abstract:

By microscopic observation and photography, the morphological characteristics and distribution of chromatophores during the early life history of Pterapogon kauderni were investigated, as well as the body color changes. The results showed that it took 15~22 days for a fertilized egg to hatch under (27±0.5) ℃. The embryo developed to the organ formation stage on the 4th day post fertilization (dpf). Melanin appeared in the eyeball at 6 dpf, xanthophore appeared at the somites at 7 dpf, reflecting platelets appeared in the eye at 8 dpf, and leucophores emerged on the second dorsal fin at 19 dpf. The formation of body color pattern of P. kauderni began with the appearance of melanocytes in the interorbital space at 12 dpf; the first pigment band in the snout became visible at 21 dpf; the yolk sac was completely absorbed and the four lateral pigment bands on the body and the Y-shaped pigment band at the tail were largely formed on the 1st day post hatching (dph). White spots started to appear between the third and fourth pigment bands in the middle of the body at 70 dph, and the body color was consistent with that of a 2-year-old adult fish at 220 dph. The research results can provide a theoretical basis for exploring the regulation mechanism of body color formation in P. kauderni and the development of ornamental fish evolution theory, as well as their genetic breeding.
