Cytotoxic steroids from the soft coral Sinularia flexibilis collected off the Xisha

摘要:

文章对采集自我国南海西沙群岛的短指软珊瑚Sinularia flexibilis的化学成分及其生物活性进行研究。采用薄层层析、硅胶柱层析、凝胶柱层析和半制备高效液相色谱等各种分离方法进行分离纯化, 根据理化特性和各种波谱数据, 结合文献数据对分离到的化合物进行结构鉴定。从短指软珊瑚S. flexibilis中分离鉴定了8个甾体类化合物, 其结构分别鉴定为: 24-亚甲基胆甾-5,24(28)-二烯-3β,19-二醇(24-methylcholesta-5,24(28)-diene-3β,19-diol) (1)、(3β,23S)-ergosta-5,24(28)-diene-3,23-diol (2)、4α-methyl-3β,14β-dihydroxy-5α-ergost-24(28)-en-23-one (3)、sinulasterol C (4)、nephalsterol C (5)、24-亚甲基胆固醇(24-methylene-cholesterol) (6)、4α-methyl-3β,8β-dihydroxy-5α-ergost-24(28)-en-23-one (7)和4α-甲基-麦角甾-24-烯-3β-醇(4α-methyl-ergosterol-24-en-3β-ol) (8)。肿瘤细胞毒活性测试结果表明, 化合物235具有较强的K562 (人慢性髓原白血病细胞)毒活性, 其IC50 (半数最大抑制浓度)值分别为(5.48±0.22)μmol·L-1、(4.80±0.11)μmol·L-1和(2.08±0.11)μmol·L-1

Abstract:

Thin layer chromatography, silica gel column chromatography, gel column chromatography and semi preparative high-performance liquid chromatography were used to study the chemical compositions of Sinularia flexibilis, a soft coral collected off the Xisha islands in the South China Sea. Eight steroids were identified from the soft coral S. flexibilis, 24-methylcholesta-5,24(28)-diene-3β,19-diol (1), (3β,23S)-ergosta-5,24(28)-diene-3,23-diol (2), 4α-methyl-3β,14β-dihydroxy-5α-ergost-24(28)-en-23-one (3), sinulasterol C (4), nephalsterol C (5), 24-methylene-cholesterol (6), 4α-methyl-3β, 8β-dihydroxy-5α-ergost-24(28)-en-23-one (7) and 4α-methyl-ergosterol-24-en-3β-ol (8) by the physical and chemical properties, extensive spectroscopic analysis and comparison with the reported data from literatures. The results of cytotoxic activity test showed compounds 2, 3 and 5 had strong inhibitory activity on K562 (human chronic myeloid leukemia cells), with IC50 (half maximal inhibitory concentration) values of (5.48 ± 0.22) μmol·L-1, (4.80 ± 0.11) μmol·L-1 and (2.08 ± 0.11) μmol·L-1, respectively.
