广西红树林放线菌 Streptomyces olivaceuss 10127 次级代谢产物及降糖降脂活性研究

摘要:

红树林生态环境特殊, 具有丰富的放线菌资源, 是天然活性物质的来源之一。为获得更多具有良好生物活性的化合物, 对一株来源于广西茅尾海红树林老鼠簕(Acanthus ilicifolius)根际土壤的链霉菌属放线菌Streptomyces olivaceuss 10127的次级代谢产物以及降糖降脂活性进行研究。采用硅胶柱色谱、高效液相色谱和半制备液相色谱等技术, 对该菌株的次级代谢产物进行分离。通过PCR扩增和16S rRNA基因测序对菌株进行鉴定, 借助核磁共振(nuclear magnetic resonance, NMR)、质谱(mass spectra, MS)以及旋光(optical rotatory dispersion, ORD)等波谱学方法和文献数据对单体化合物进行结构鉴定。通过试剂盒测定化合物的降糖降脂活性。从Streptomyces olivaceuss 10127的次级代谢产物中共分离得到12个单体化合物, 分别为: (6-(3-methylbut-2-enyl)-1-indol-3-yl)methanethiol(1)、methyl indole-3-acetate(2)、methyl 4-hydroxyphenylacetate(3)、2-hydroxybenzamide(4)、germicidin B(5)、germicidin A(6)、isogermicidin A(7)、4′-hydroxy-5, 7-dimethoxy-6-(3-methyl-2-butenyl)-isoflavone(8)、2, 3-dihydroxybenzoic acid(9)、phenyl acetic acid(10)、germicidin N(11)、piloquinonea(12)。其中, 化合物812为首次从该放线菌中分离得到。首次测定了分离得到的化合物的降糖降脂活性。化合物1在降低脂质方面表现出较好的效果; 化合物7不仅具有降脂效果, 还具有降糖效果, 且效果均优于阳性药组。放线菌Streptomyces olivaceuss 10127具有生产降糖降脂活性先导化合物的潜在研究价值。

Abstract:

Mangrove has a special ecological environment and abundant actinomycetes resources, which is one of the sources of natural active substances. To obtain compounds with potent biological activity, this study was conducted on the secondary metabolites and hypoglycemic and lipid lowering activities of an actinomyces, Streptomyces olivaceuss 10127. The strain was derived from the rhizosphere soil of mangrove Acanthus ilicifolius L. in Guangxi. The strains were identified by PCR amplification and 16S rRNA gene sequencing, and the secondary metabolites of this strain were isolated by silica gel column, high performance liquid chromatography (HPLC) and semi-preparative liquid chromatography. The structures of monomer compounds were identified using nuclear magnetic resonance (NMR), mass spectra (MS) and optical rotatory dispersion (ORD). The hypoglycemic and lipid-lowering activity of the compound was tested by the kit. From the secondary metabolites of Streptomyces olivaceuss 10127, 12 compounds were isolated. These include (6-(3-methylbut-2-enyl)-H-indol-3-yl) methanethiol (1), methyl indole-3-acetate (2), methyl 4-hydroxyphenylacetate (3), 2-hydroxybenzamide (4), germicidin B (5), germicidin A (6), isogermicidin A (7), 4′-hydroxy-5, 7-dimethoxy-6-(3-methyl-2-butenyl)-isoflavone (8), 2, 3-dihydroxybenzoic acid (9), phenyl acetic acid (10), germicidin N (11), piloquinonea (12). Compounds 8 and 12 were isolated from this actinomyces for the first time. The hypoglycemic and lipid-lowering activity of the isolated compounds were assayed for the first time. Compound 1 showed a good effect on lipid reduction. Compound 7 showed both the lipid lowering effect and the glucose lowering effect, surpassing the efficacy of the positive control group. The actinomyces Streptomyces olivaceuss 10127 has potential research value in producing lead compounds with hypoglycemic and lipid lowering activities.
