Research progress of the regulating hormones for spawning behaviors of sea cucumbers

Abstract

Sea Cucumbers play an important role in marine ecological system and have edible and medicinal values. At present, wild resources of sea cucumbers are greatly depleted in China, and artificial breeding of sea cucumbers provides an essential way to restore the population resources. Understanding the hormones that regulate spawning behaviors of sea cucumbers has great significance for the development of artificial injection hormone. We reviewed the hormones found in sea cucumbers that can regulate their spawning behaviors, and we also provide some new perceptions for the future research. The review can provide some valuable references for revealing the key spawning mechanism of sea cucumbers and developing the artificial spawning induction by injecting hormones.

海参隶属于棘皮动物门(Echinodermata)海参纲(Holothuroidea), 是著名的“海洋清道夫”, 具有重要的生态功能, 是海洋生态系统平衡的重要维持者(Purcell et al, 2016; Conand, 2018)。同时, 海参也是“海产八珍”之一, 有相当一部分海参具有重要的食用或药用价值(张春云 等, 2004; 邹峥嵘 等, 2004)。中国以及东亚国家是海参的主要消费市场, 近30年来由于本地区经济发展和消费水平的提高, 海参消费需求强劲, 直接刺激了世界海参捕捞业的扩张。据统计, 每年有近10万t野生海参被捕捞, 约70%的野生热带海参种群资源受到过度捕捞威胁, 导致世界野生海参资源严重受损(González-Wangüemert et al, 2018)。因此, 进行大规模人工繁育海参、培育优质苗种迫在眉睫, 一方面可以用于增殖放流、修复海参种群资源, 另一方面可以用于人工养殖, 满足人们对海参的消费需求, 减少对野生海参资源的依赖。
在海参的人工繁育中, 通常采用阴干流水法进行海参的人工催产, 但是这种方法具有亲参需求量大、雌参排卵比例低、产卵时机不易掌控的不利因素, 严重制约了海参增养殖业的发展以及海参种群修复工作。类似于鱼类催产, 开发出海参的人工注射催产激素将为大规模、稳定、低成本的海参苗种人工繁育作出里程碑式贡献。挖掘调控海参产卵行为的各种激素, 有助于阐明海参产卵的激素调控机制, 将为开发高效的海参人工催产药剂提供关键理论支撑。

1 海参产卵行为的调控激素

20世纪80年代, 研究者们已经发现海参产卵行为受到激素的调控。1985年, Maruyama (1985)首次发现来自5种不同海参的桡神经提取物(radial nerve extract, RNE)可促进4种受试海参的离体性腺片段卵母细胞成熟, RNE催熟海星性腺表现出物种交叉性。后续研究发现海星的RNE同样也促进新西兰海参(Australostichopus mollis)卵母细胞成熟, 表明刺激性腺成熟因子在棘皮动物中可能具有高度的保守性(Maruyama, 1986)。研究者试图从海参RNE中纯化并鉴定起关键作用的多种激素分子, 这对于解析海参性腺成熟、产卵的激素调控机制也具有重要价值。

1.1 松弛素样性腺刺激肽

关于海参松弛素样性腺刺激肽(relaxin-like gonad-stimulating peptides, RGP)的研究起步非常晚, 相关资料也非常少, 但可以从海星RGP的研究中得到启示。1959年, Chaet等(1959)首次发现福氏海盘车(Asterias forbesi)桡神经索的提取物注射能够诱导其产卵, 后续研究中将这种能够刺激性腺发育和产卵的物质命名为GSS (gonad-stimulating substances)。2009年, Mita等(2009)首次从海燕(Asterias pectinifera)的桡神经索中纯化了GSS, 并鉴定GSS为一种松弛素样的肽, 因此GSS被重新命名为松弛素样性腺刺激肽(RGP)。此后在多棘海盘车(A. amurensis)和日本滑海盘车(Aphelasterias japonica)中也发现了同源的RGP (Mita, 2016)。成熟的RGP为一种异源二聚体肽, 长度为43aa, 包括A链和B链, A链和B链通过中间的二硫键进行交联, A链具有松弛素超级家族典型的半胱氨酸基序(C基序)(Mita, 2021), 只有维持正确四级结构RGP才具有活性(Chieu et al, 2019a)。RGP前体蛋白还包括了位于A链与B链之间的C-肽段, 但C-肽段在前体肽翻译后被胰蛋白酶样的蛋白酶加工切除(Mita, 2013)。在非繁殖季节, RGP仅存在于海星的神经组织, 只有在繁殖季节才可在体腔液中检测到RGP (Mita, 2021)。RGP被鉴定并明确功能后, 人工合成的RGP被用于多种海星的体内或体外催产, 并取得成功(Mita, 2021), 这些研究表明, RGP在海星的人工催产中具有很好的应用潜力。
海星RGP的研究也推动了海参RGP的研究。Chieu等(2019b)首次在糙海参中鉴定出编码RGP的基因, 通过酵母表达的重组RGP高效诱导了糙海参的产卵。同年, 他们利用生物信息学方法, 在玉足海参中鉴定出43个神经肽的前体基因, 其中有7个被认为参与了棘皮动物的繁殖, 包括RGP、Cubifrin、肌调蛋白样神经肽、促皮质素肽等(Chieu et al, 2019a)。黄乳参(Holothuria fuscogilva)是一种稀有的高价值海参, Nocillado等(2022)通过转录组数据等获得了黄乳参的RGP基因序列, 通过酵母表达的重组RGP高效诱导了黄乳参的产卵, 这为黄乳参的繁育以及种群资源恢复提供了有力的技术支撑。

1.2 性腺刺激物样分子

2009年Katow等(2009)第一次从仿刺参(Apostichopus japonicus) RNE中分离得到一种可在体外刺激卵母细胞成熟的多肽-性腺刺激物样分子(gonad-stimulating substance-like, GSSL), 并通过质谱、RACE等手段获得了GSSL编码基因cDNA序列和GSSL的全序列, GSSL全长包括43个氨基酸。人工合成的GSSL以及GSSL的N端21个氨基酸的短肽获得了与RNE同样的刺激卵母细胞成熟的效率。但是未见采用GSSL进行体内注射催产的后续报道。GSSL虽然与后期在海参中发现的RGP具有一样的肽链长度, 但GSSL与RGP完全不具有相似性, 也不具有松弛素超级家族所具有C基序, 因此GSSL并非RGP。
除了RNE能分离出GSSL外(Katow et al, 2009; Ahmed et al, 2011)发现仿刺参各个组织全年都有GSSL的表达, 并在仿刺参桡神经分离出GSSL的前肽(gonad-stimulating substance-like peptide-containing polypeptide, GSSLP), 为170kDa的单一肽, GSSLP的N端糖基化程度很低, 且表达水平全年变化不大。GSSLP主要存在于桑椹胚细胞中, 由这些细胞转运到体腔上皮的边缘。不同于桡神经, 性腺组织的GSSLP丰度与繁殖季节具有相关性, 例如: 3—6月, GSSLP在仿刺参卵巢的卵泡细胞中有较多表达, 而在7月, GSSLP表达量较少(Ahmed et al, 2011)。此外, 处于繁殖季节的仿刺参卵巢、精巢中的GSSLP分子构象呈现出多态性, 这种现象被解释为性腺不仅仅从体腔液接收GSSLP, 也能在桑椹胚细胞中通过N端糖基化和二硫键修饰GSSLP, 产生组织特异的GSSLP (Ahmed et al, 2011)。根据生殖腺中GSSL表达的季节性, Katow等(2014)研发了一种GSSL免疫层析检测试剂盒(GIM-kit), 能够简单快速检测海参卵巢是否成熟, 提高了海参人工催产的效率。

1.3 五肽酰胺

Kato等(2009)用仿刺参口环神经提取物以及RNE与海参卵巢片段共同孵育, 诱导了卵巢片段中卵母细胞成熟和排卵, 从该提取物中进一步纯化获得了一个五肽和一个七肽, 注射两种短肽均能诱导海参卵母细胞成熟和产卵释放, 七肽的诱导效果稍差。五肽和七肽分别被鉴定为 NGIWY五肽酰胺(NGIWYamide)、QGLFSGV七肽酰胺(QGLFSGVamide)。此外, 还发现人工合成的NGLWYamide (第三位氨基酸改为亮氨酸)的诱导产卵的效率比天然的NGIWYamide提高10 ~ 100倍(Kato et al, 2009), NGLWYamide注射后大约2h, 雌、雄仿刺参亲本达到同步产卵或排精(Fujiwara et al, 2010)。NGLWYamide活性依赖五肽C-末端的酰胺, NGIWYamide及其衍生物NGLWYamide分别被命名为Cubifrin-I和Cubifrin-L (Fujiwara et al, 2010)。Cubifrin-L被认为是一种非常有潜力的仿刺参注射催产药剂(Katow et al, 2009; Fujiwara et al, 2010)。宋坚等(2009)通过人工催产实践发现, Cubifrin-L单独用于仿刺参人工催产并未取得理想效果, 采用Cubifrin-L与氯化钾(KCl)组合的药剂进行仿刺参的人工催产, 取得了比单独使用Cubifrin-L更好的催产效果。
陈廷等(2019)从小疣刺参(Stichopus monotuberculatus)中鉴定出Cubifrin的前体基因, 发现其编码3种短肽: Cubifrin-I、Cubifrin-L、NGFWYamide, 注射催产发现3种五肽均可刺激小疣刺参产卵, 但NGFWYamide的效果明显优于另外2种五肽。Chaiyamoon等(2020)通过生物信息学的方法, 鉴定出糙海参(Holothuria scabra) Cubifrin的前体基因, 其成熟的编码产物不仅包括Cubifrin-I还包括NGIWFamide (Cubifrin-F), Cubifrin-F刺激糙海参产卵效率达到70%, 远高于Cubifrin-I (10%)。Cubifrin的前体基因在糙海参卵巢发育后期卵巢管壁高表达, 表明Cubifrin很可能通过引起糙海参卵巢管壁收缩的方式刺激产卵(Chaiyamoon et al, 2020)。

1.4 成熟促进因子

1970年Stevens (1970)发现在1-甲基腺嘌呤(1-MeAde)的处理下, 海星卵母细胞内产生一种成熟促进因子(maturation promoting factor, MPF), MPF促成了卵母细胞生发囊泡破裂(germinal vesicle breakdown, GVBD)以及后继的配子发生过程。Kishimoto等(1982)发现将含有MPF的成熟海星卵母细胞质注入未成熟海参卵母细胞中, 可以诱导海参卵母细胞GVBD。海星MPF被鉴定为一种异源二聚体激酶, 包括单分子cdc2多肽和细胞周期蛋白B (Labbé et al, 1989)。由于MPF仅在细胞质内发挥作用, 因此不能直接用于人工催产。

1.5 其他未知调控激素

Chieu等(2018)通过更容易操作的提取方式获得了玉足海参(Holothuria leucospilota)的RNE, 发现玉足海参RNE可诱导其卵母细胞成熟, 效率高达98.9%, NRE催熟所产卵子99%可以正常受精并正常发育, 表明NRE具有在海参人工催产实践中使用的潜力。玉足海参RNE中具有与仿刺参同样的NGIWYamide, 但是合成的NGIWYamide并不能诱导玉足海参卵母细胞成熟, 这一结果表明RNE中存在其他激素对玉足海参的性腺成熟起关键作用(Chieu et al, 2018)。

2 1-甲基腺嘌呤及其类似物在海参产卵中的作用

1-MeAde是无脊椎动物中最早发现的生殖调控激素, 可以诱导海星卵母细胞的成熟以及海星的产卵(Stevens, 1970; Kubota et al, 1977)。此外, 一些常见的二硫化物还原剂(dithiothreitol, DTT)、二巯基丙醇(2,3-dimercapto-l-propano, BAL)、L-半胱氨酸(l-cysteine, L-cyst)发挥了类似1-MeAde作用, 也可诱导海星卵母细胞成熟及排卵(Kishimoto et al, 1980)。在实际应用方面, 1-MeAde被成功用于棘冠海星(Acanthaster planci)的高效人工催产, 人工催产的幼体完全可以正常发育成稚海星(Keesing et al, 1997; Tian et al, 2017)。
鉴于1-MeAde及其类似物在海星卵母细胞成熟中的作用已经明确(见本文第3部分), 1-MeAde及其类似物最早被用于探索在海参卵母细胞成熟以及诱导产卵中的作用。结果表明, 它们在不同海参中作用表现并不一致。1-MeAde可刺激粘细海参(Leptosynapta inhaerens)离体卵巢片段中卵母细胞的最终成熟及排卵(Ikegami et al, 1976), 1-MeAde也可使40%的糙海参离体卵母细胞成熟(Léonet et al, 2009)。然而采用类似的方法, 1-MeAde并不能诱导加利福尼亚海参(Stichopus californicus)(Stevens, 1970)和仿刺参(Kishimoto et al, 1980)卵母细胞的成熟及排卵。1-MeAde类似物DTT、BAL、L-cyst 可刺激玉足海参、豹纹海参(H. pardalis)离体卵母细胞100%成熟并排卵(Maruyama, 1980), 但是在糙海参中, 3种类似物诱导卵母细胞成熟的效率分别为100%、80%、40% (Léonet et al, 2009)。1-MeAde、DTT、BAL、L-cyst也被用于诱导叶瓜参(Cucumaria frondosa)卵母细胞成熟, 但只有DTT诱导卵母细胞成熟, 且排卵的比例最高(48%)(Gianasi et al, 2019)。1-MeAde及其类似物诱导不同海参卵母成熟的差异结果, 提示海参中卵巢卵泡细胞产生第二介质很可能是一种类似于1-MeAde的嘌呤衍生物, 海参中的卵巢催熟第二介质究竟是什么, 还需要进一步研究。

3 海参产卵激素的调控机制

由于海参具有重要生态功能和经济价值, 研究者更倾向于直接挖掘或筛选与海星产卵调控激素类似的激素, 并尝试应用于海参催产, 这可能是导致海参产卵激素调控机制研究领域相对空白的重要原因(Léonet et al, 2009)。截至目前, 海星产卵的激素调控机制已经基本被阐明, 海星、海参同属棘皮动物, 一般认为, 棘皮动物门卵母细胞成熟的过程相似, 不同物种之间差别较小。因此, 海星产卵的激素调控机制对于海参相关研究具有重要的参考价值。
海星的产卵行为受到神经肽类激素的严密调控(Mita, 2013), 其激素调控的关键机制见图1。海星的桡神经索内存在RGP (Mita et al, 2009; Mita, 2013; Mita, 2016), RGP由桡神经索分泌, 通过体腔液运输到性腺, 作用于卵巢卵泡细胞, RGP的受体属于视紫红质样G蛋白耦联样受体超级家族。RGP受体主要与Gs蛋白耦联, RGP控制卵巢卵泡细胞及睾丸间质细胞中环磷酸腺苷(cyclic adenosine monophosphatec, cAMP)和1-MeAde的产生(Mita, 2013; Mita, 2016; Mita, 2021)。调节RGP生产的机制和因子未知, 目前已知RGP由离子霉素诱导后在桡神经索分泌, RGP是一种钙离子载体, 因此认为细胞内钙离子浓度增加可以介导RGP的刺激分泌(Mita, 2021)。Lin等(2017)发现海燕分泌RGP的神经元细胞与体表层感觉器官细胞紧密相连, 表明感觉器官细胞很可能是外界环境因子与神经激素调控产卵行为的中介者。1-MeAde作为第二介质作用于卵母细胞膜受体以激活细胞内MPF (Stevens, 1970; Yamashita et al, 2000), 激活的MPF诱导卵母细胞的成熟, 卵母细胞成熟的过程包括了GVBD、染色质浓缩和极体外排(Léonet et al, 2009)。

4 研究展望

海参产卵的激素调控研究较少, 起步较晚, 现代分子生物技术和测序技术的发展和普及, 将大大加快我们对海参产卵激素调控机制的研究, 最终揭示出海参产卵的激素调控机制。未来相关研究应当从以下几方面开展:
1) 开展RGP、五肽等对海参产卵促进作用相对明确的产卵激素高效表达或人工合成研究, 开发效果稳定的商业化催产激素产品, 并在海参人工繁殖生产中应用, 从而提高海参人工繁殖技术水平。
2) 在海参的人工繁殖中提前掌握海参成熟程度有利于精准筛选出优质亲参, 减少亲参的用量, 并提高繁育工作的效率, 因此开发适用于我国经济海参种类的性腺成熟度检测试剂盒产品具有重要的实践意义。
3) 进一步挖掘其他对海参产卵起关键作用的激素, 开发成为高效的海参人工催产药剂, 用于海参的大规模繁育, 进而推动海参增养殖产业的发展以及海参种群资源的恢复。
4) 综合应用现代分子生物技术深入揭示海参产卵激素的调控机制。
