Effects of nitrate and phosphate on the growth and toxin production of Amphidinium klebsii (Sanya strain)

Abstract

In order to evaluate the growth and toxin production of Amphidinium klebsii (Sanya strain) under different nitrate and phosphate concentrations, a series of nitrate and phosphate solutions were prepared based on modified K medium. Nitrate and phosphate had significant effects on the growth and toxin productivity of A. klebsii. Low-nitrate and low-phosphate concentrations (N concentration≤4.665×10-4mol•L-1, P concentration≤2.000×10-5mol•L-1) in culture media were not favorable to the biomass accumulation of A. klebsii but favorable to its growth and toxin production. On the contrary, middle-nitrate and middle-phosphate concentrations (4.665×10-4mol•L-1-4mol•L-1, 2.000×10-5mol•L-1-5mol•L-1) were favorable to the biomass accumulation of A. klebsii but not favorable to its toxin production. Extra-high-level nitrate and phosphate concentrations (N concentration≥37.320×10-4mol•L-1, P concentration≥16.000×10-5mol•L-1) seemed to inhibit the growth of A. klebsii, but resulted in higher cell densities and contributed to its toxin production. Under the experimental condition, optimum growth N and P concentrations were 4.665×10-4mol•L-1-4mol•L-1, 2.000×10-5mol•L-1-5mol•L-1. The highest biomass value estimated in culture was 1.237×105mL-1 at the levels of nitrate 9.330×10–4mol•L–1 and phosphate 4.000×10–5mol•L-1, and the maximum hemolytic activity reached 347.48HU•L-1 under the same condition.
